Long-Term Contraceptive Use Linked to Higher Brain Tumor Risk, Study Finds
A major new study has raised significant concerns about the long-term use of hormonal contraceptives, suggesting a link between certain birth control pills and injections and an increased risk of developing a specific type of brain tumor. Researchers in Denmark analyzed decades of medical records and found that women using these methods faced a notably higher chance of being diagnosed with meningioma, a tumor that forms in the membranes surrounding the brain and spinal cord.
While the word “tumor” often triggers fear of cancer, experts clarify that most meningiomas are non-cancerous. What the Study Revealed About Contraceptive Risks
The research, which examined the medical records of three million women over 25 years, focused on the link between hormonal contraceptives and meningioma. Scientists compared 1,473 women who had been diagnosed with the tumor against 14,717 women without it. The findings were striking.
The Highest Risk: Injectable Contraceptives
The strongest association was found with the injectable contraceptive medroxyprogesterone, sold under the brand name Depo-Provera in many countries. Women using this injection had a 355 percent higher risk of developing meningioma compared to those who did not use it.
Age played a significant role in the level of risk. For women aged 55 to 59 who used the injection for one year, researchers estimated that one additional case of meningioma would occur for every 5,372 women. In contrast, the risk was much lower for younger women. Among those aged 15 to 19, only one extra case was predicted per 449,000 users.
Combination Pills Also Show Elevated Risk
The study, published in the JAMA Network journal, also examined combination birth control pills containing both estrogen and progestogen. Several types were associated with increased risk:
- Desogestrel: 66% higher risk
- Cyproterone: 61% higher risk
- Drospirenone: 58% higher risk
- Gestodene: 44% higher risk
- Levonorgestrel: 40% higher risk
- Norethisterone: 38% higher risk
- Norgestimate: 4% higher risk
These numbers highlight that while the risk varies depending on the specific hormone composition, it is not limited to high-dose formulations alone.

Scientists from the Danish Medicines Agency noted that previous research had linked the risk primarily to high-dose hormonal drugs and the Depo-Provera injection.
